Right, listen up you sorry lot! League of Legends is a free to play game with micro transactions which isn't a big deal. It's from the same creators of DotA and is similar to Demigod in some respects but slightly more cartoony but don't let that bother you.
To sign up, go to the European site (hint hint Redders) and register then download the client.
Once you make your account, choose your 'summoner' avatar I recommend assigning your 'Mastery' points and runes before you start. Click on 'View Summoner Profile'.
You should be presented with a summary of your stats, games played, kills, deaths etc. You should start off with one rune and one mastery point - these add permanent stat boosts to your chosen champion so it's worthwhile to assign them when you can. It'll look something like this:
All you do is drag the rune into the relevant rune category/colour. You'll get more rune slots in your rune book every time you level up and there are loads of different runes available to buy from the store. Don't worry though, cos you win 'Influence Points' every time you finish a match which you can spend on runes or new champions in the store so you needn't spend a penny.You can see how many 'IP' you have on the top right corner signified with the two swords. There are two pages in the rune book but you can only choose one per game so you can have one page set up to a different play style if you so wish to.. You can take them off so your choices aren't permanent.
Next up is the 'Masteries' section. These are also permanent stat boosts but you get a new point to spend every level you gain.
The spells section is the last bit you need to know. You have a common 'pool' of spells you can select two from before every game no matter which champion you choose.
When you start a game, either practice, or normal you will get 1:30 to select which champion you want, rune book page and your two spells. You can only have one of each champion per team so any champion already chosen by another member of your team will be greyed out.
There are two teams, each with a set of champions. The aim of the game is to destroy the enemys' nexus which is right in the heart of their base. Of course there are powerful towers along the way which are quite deadly at low levels, plus the waves of minions that come out of the nexus frequently. Then you have the enemy champions themselves which will be tasked with destroying your nexus.
Each champion has only four skills you can level up so the main way you can get an edge on the enemy is to buy items from the store. You have six item slots and the more expensive items are actually a combination of the cheaper items. There are a lot of items so it'll take a while to see what items you need/want but there is an online item database on the link I posted above. You get gold from killing the little minions and enemy champions but also your champion generates gold passively as well. You can buy items which further boosts this passive gold ability - 'Heart of Gold' being one of them.
For those starting out you have a limited number of champions to choose from out of the 48 available. You can buy those you want with your 'Influence Points' or use real money to buy 'Combat Points' (which are signified with a red fist above your 'Influence Points'), up to you. Every week they have 10 different champions for you to play with for free to try them out so it's not too bad. What I'd recommend is that you just pop into Game and buy the game for £4.99, this will not only give you a code to unlock loads of champions off the bat (much cheaper than buying them with real money) but also give you a bit of 'Combat Points' to buy extra champions/skins with.

As you can see there are two prices for champions etc. One is the 'Combat Point' cost and the other 'Influence Point' cost, you'll notice that the 'IP' cost is higher than the 'CP' since it's free to get (you just need to play lots). It should give you an idea of how the micro-transaction system works. Runes, thankfully, only cost 'IP' so feel free to spend away!
Anyway hope this helps explain how to set up and configure your summoner on League of Legends. It's a pretty good game especially since you don't need to spend any money playing it.
